摘要
通过免疫荧光共定位实验,研究了RNA聚合酶Ⅱ的亚基Rpb1,Rpb6,Rpb8与BAF复合物和转录因子NF1/CTF在基因转录活动中的联系.实验证实,RNA聚合酶Ⅱ的亚基Rpb1与BAF复合物有联系,Rpb6与BAF复合物联系不紧密,Rpb6与NF1/CTF联系密切,Rpb8与BAF复合物和NF1/CTF有部分联系.免疫沉淀实验也证实RNA聚合酶Ⅱ与BAF复合物和NF1/CTF一同参加了转录起始复合物的形成.
This paper providest the evidences that the subunit Rpbl, Rpb6, Rpb8 of RNA polymerase Ⅱ have relationship with BAF complex by Immunofluorescene co - orientation assay . Tests validate the sub- unit Rpbl of RNA polymerase Ⅱ have relationship with BAF complex, the small subunit Rpb6 of RNA polymerase Ⅱ have no obvious orientation relationship with BAF complex, then the small subunit Rpb6 of RNA polymerase Ⅱ has obvious orientation relationship with transcription factor NF1/CTF, the small subunit Rpb8 of RNA polymerase Ⅱ have a little orientation relationship with BAF complex and NF1/CTF. The results of Immunoprecipitation assay further demmonstrates RNA polyrnerase Ⅱ and BAF complex with transcription factor NF1/CTF take participate in formation of transcriptional initiation complex together.
