宫颈癌组织高危HPV18 L1基因的克隆及序列分析 被引量:5

摘要 目的克隆和分析人乳头瘤病毒18型晚期表达基因L1序列,为HPV感染的检测和基因工程疫苗研制提供基础。方法采用PCR技术扩增了1例HPV18阳性宫颈腺癌患者癌组织中HPV18L1基因,并对其进行了克隆及全序列分析。结果本例宫颈癌临床标本HPV18L1基因与GenBank收录的原型比较有12个碱基变异,推导其编码的氨基酸也发生了变化。结论本例中国人宫颈癌HPV18L1基因有一定的变异。 Objective To clone and analyze the whole sequence of L1 gene and its deduced L1 protein of HPV18 from specimen of cervical cancer. Methods HPV18 LI gene was amplified from a HPV18-positire DNA sample of a Chinese patient with cervical ad by means of PIER and then cloned and sequenced. Results In comparison with the prototype, there were twelve point mutations in HPV 18 L1 nucleic acid sequence and the differed sequence had changed the triplet codes. Therefore, the amino acid sequence of HPV 18 L1 protein could be subsequently changed. Conclusion Some mutations had taken place in the nucleotide sequence of HPV18 L1 gene obtained from the cervical cancer tissue of this Chinese patient.
