摘要
目的建立高效液相色谱-蒸发光散射检测法测定脂肪乳注射液中磷脂酰胆碱（PC）含量。方法采用Lichrospher 100Diol柱（4．6mm×250mm，5μm），以正庚烷-异丙醇（86：114）为流动相A，正庚烷-异丙醇-水（31：62：12）为流动相B，进行梯度洗脱。流速为1．0mL·min^-1，柱温为40℃；蒸发器温度80℃，空气，喷气流速为2．0L·min^-1，结果磷脂酰胆碱的线性范围为0．134～0．938g·L^-1，r=0．9982（n=7），PC加样回收率为98．9％，RSD为0．3％（n=9）。结论本法操作简便快速、重复性好、灵敏度高、准确、专属性高。
OBJECTIVE To develop a HPLC-ELSD method for the de.termination of phnsphatidyl choline（PC） content in the medium and long chain fat emulsion injection. METHODS The separation was performed on a Lichrospher 100 Diol cnhann （4.6 mm×250 mm, 5μm） with n-heptane-iso-propanol（86： 114） as mobile phase A and n-heptane-iso-propanol-water（31：62：12） as mobile phase B using linear gradient elution. The flow rate was set at 1.0 mL·min^-1. Column temperature was 40℃, inlet heater was 80℃. The air flow rate was 2.0 L·min^-1.RESULTS The linear range for the content of PC was 0.134～0.938 g·L^-1,r=0.9982（n=7）. The recovery of PC was 98.9% and its RSD was 0.3% （ n = 9）. CONCLUSION The method is simple, rapid, reproducible, good sensitive, accurate and is suitable for the determination of phosphatidyl choline.
