摘要
提取绵羊睾丸总RNA,并以此为模板,采用RT-PCR技术扩增出精子表面特异性蛋白PH-20的cDNA。应用T/A克隆策略,将扩增的PH-20基因克隆入T载体,通过酶切和测序进行鉴定,结果表明,PH-20 mRNA在绵羊睾丸和附睾中均有表达。
The sperm surface specific protein PH-20 is a single-chain protein, anchored in the plasma membrane with a glycosyl phosphatidylinositol （GPI） linkage and localized on the plasma membrane of the posterior sperm head region and inner acrosomal membrane. It has been shown to possess hyaluronidase activity, which is play an important role in the fertilization process. In this study, total RNA was prepared from sheep testis, cDNA fragment was amplified by RT-PCR using specific primers, and then was cloned into pGM-T Easy vector. Inserted PH-20 gene was sequenced. Thus,the PH-20 gene was proved to be expressed in both the testis and epididymis of sheep.
