摘要
目的以实时定量聚合酶链反应(PCR)测定三叉神经痛大鼠延髓内降钙素基因相关肽(CGRP)量的变化,探讨CGRP与三叉神经痛发病机制的关系。方法12只SD大鼠随机均分为4组。A、C组为手术组,用铬线疏松结扎大鼠右侧眶下神经造成慢性缩窄性损伤;B、D组为假手术组,仅暴露神经,不结扎。观察术前及术后3、6、9、12、15d大鼠对机械性疼痛刺激的反应阈值。A、B组术后9d及C、D组术后15d分别取大鼠延髓组织,应用实时定量PCR法测定CGRP的量。结果与假手术组比较,手术组大鼠术后9~15d右眶下神经支配区域机械性痛觉超敏(P〈0.05)。A组大鼠延髓内CGRP量明显多于B组(P〈0.05),C组大鼠延髓内CGRP量明显多于D组(P〈0.05)。结论眶下神经的慢性环扎损伤可导致大鼠三叉神经痛,且大鼠疼痛时CGRP的表达增高。
Objective To assess the relationship between calcitonin gene-related peptide (CC-RP) in medulla oblongata of rats' trlgeminal neuralgia by using real-time polymerase chain reaction (PCR). Methods Twelve SD rats were divided into four groups randomly, with three rats for each. Group A and C were surgical groups, Group B and D were sham operation groups. In the surgical group, right unilateral chronic constricting injury (CCI) of the rat was produced by placing loose chromic gut ligature around the infraorbital nerve (ION). In the sham operation group, the ION was exposed only using the same procedure but not ligated. Mechanical response threshold was observed before operation and 3, 6, 9, 12, 15 days after operation. All members of medulla oblongata of rats in group A and B were taken to measure the quantity of CGRP by real-time PCR nine days after operation, and those in group C and D were similarly done fifteen days after operation. Results Compared with the sham operation group, changes of allodynia to mechanical stimulation on the territory of ligated ION were found from the 9th to 15th day after operation in the surgical group (P〈0.05). The expressions of CC-RP in medulla oblongata of group A were greater than those in group B (P〈0.05) and the expressions of CGRP in medulla oblongata of group C were also greater than those in group D (P〈0.05). Conclusions CCI-ION can result in trigeminal neuralgia of rats with increas expression of CGRP in neuralgic rats. (Shanghai Med J, 2006, 29 : 101-103)
