摘要
哺乳动物膀胱上皮中存在一种独特的不对称单位膜(AUM)结构。本文应用多种电镜技术,观察了小鼠和牛的AUM的形态结构,并对AUM蛋白及其mRNA进行定性与定位研究,在此基础上探讨了膀胱上皮细胞分化中AUM结构与成分的变化,结果显示:(1)小鼠膀胱上皮中间层细胞开始转录AUM的mRNA并翻译AUM蛋白形成典型的AUM结构,而牛只有在表层细胞中表达AUM蛋白,它们可能代表两种不同类型的膀胱上皮细胞的终末分化形式。(2)14天胎鼠膀胱上皮细胞中已有AUM蛋白表达,在整个发育过程中伴随AUM蛋白的表达,AUM的结构与梭形泡以及细胞表面糖被发生一系列相应的变化。(3)AUM结构与中间纤维体系紧密相连,这一结果为解释AUM的功能提供了直观的实验证据。
A unique structure of Asymmetrical Unit Membrane (AUM) exists in mammalian bladder epithelium (urothelium). The AUM characteristics and the distribution of AUM proteins (uroplakins) along with their mRNA have been examined by means of electron microscopic technique. The results demonstrate that: (1) the uroplakins and their mRNA could be detected in intermediate cells of mouse urothelium, but in bovine urothelium both of them exist only in superficial cells. It maybe represents two pathways of terminal differentiation of urothelium. (2) Uroplakins began to express in bladder urothelium of fetal mouse 14 days old. Along with the fetal development, the uroplakin expression, AUM structure and cell coat changed correspondingly. (3) AUM structure is associated tightly with intermediate filament. This result provides direct evidence for the AUM function.
