锯缘青蟹血细胞的形态及分类 被引量:17

Classification and morphological observation on haemocytes in mud crab Scylla serrata by light and electronic microscope

摘要 对锯缘青蟹(Scylla serrata)血细胞染色的抗凝剂、染色方法进行筛选。蟹样品体质量约250 g。采用亚甲基蓝、瑞氏法染色后,在Olympus油镜下观察、记数、测量,再结合电镜超薄切片观察结果对锯缘青蟹血细胞进行分类。根据血细胞质中颗粒的有无、大小、折光性、染色特性及细胞的大小、核质比等,将锯缘青蟹血细胞分为4种:(1)无颗粒细胞,细胞质中无颗粒;(2)小颗粒细胞,细胞质中有深蓝色小颗粒;(3)中间型细胞,细胞质中既有深蓝色小颗粒,又有折光性红色大颗粒;(4)大颗粒细胞,细胞质中充满了具有折光性的红色大颗粒。4种血细胞的大小顺序从小到大依次为无颗粒细胞、小颗粒细胞、中间型颗粒细胞、大颗粒细胞;核质比则相反,分别为54.01%、37.13%、25.37%、17.49%;其数量百分比分别占20.92%、40.30%、19.39%、19.39%。根据伪足的多少,对4种血细胞在机体的免疫防御机制中所起的不同作用进行了简单探讨。 The mud crab Scylla serrata is a species of well-cultured crab for its daintiness and nutritions. But the diseases frequently happened with the expanded scale of culture. This paper reported the haemocytes of Scylla serrata and its classification by selecting a simple and reliable way. The purpose of this study is to provide basic data for immunological research of Scylla serrata. By carefully observation under light and electronic microscopies, four kinds of haemocytes were classified according to their haemocytes size, morphology, nuclear size and granular existence, granular refraction, staining characters: (1) agranular hemocyte(AG) contains no granular, and a few mitochondria and endoplasmic reticulum, usually round or oblong; (2) small granular hemocyte (SG) is oblong, usually pyriform for the pseudopods, in the cytoplasm of which there are small size and blue-stainned granulars and profuse mitochondria and endoplasmic reticulum; (3) intermediate granular hemocyte (IG) is usually oblong or round, having both small and large granulars and red-stained large granulars and profusing mitochondria and endoplasmic reticulum; (4) large granular hemocyte (LG) is usually oblong, containng refractile and red-stained large granulars. In the hemocytes, the number rates of AG, SG, IG and LG are 20.92 %, 40.30 %, 19.39 % and 19.39 %, respectively. The rate of nucleus to cytoplasm (N/C) from big to small is AG (54.01% ), SG (37.13 % ), IG(25.37 % ) and LG ( 17.49 % ), but the size was contrary that they are (6.0-9.5)μm×(7.5-11.0)μm, (7.0-12.0)μm×(8.5-15)μm,(7.5 13.0)μm× (10.0-16.5)μm and (7.5 - 12.5)μm×(12.0 - 17.5)μm, respectively. Different haemocytes have different functions in the crustacean immune response based on the numbers of the pseudopods.
