分子生态技术在污染土壤微生物生态研究中的应用

摘要 分子生态学技术克服了传统微生物培养技术的限制,可以更精确地揭示环境中微生物种群结构及其遗传的多样性,使得污染微生物生态学的研究进入一个新的阶段.本文主要介绍了基于16S rDNA的分子分析技术,包括16S rD-NA序列分析、ARDRA((扩增rDNA限制性分析)、T-RFLP、RFLP(末端限制性片段长度多态性)技术、RISA(核糖体基因间隔序列分析)法、SSCP(单链构象多态性)分析、DGGE/TGGE(变性/温度梯度凝胶电泳)技术、RAPD(随机扩增多态性DNA分子标记)技术,及其在污染环境微生物多样性研究中的应用研究进展,为污染微生物生态学的进一步研究应用提供帮助. Molecular ecological techniques resolve lots of difficulties in studying the microorganisms which could not be cultured. The research of pollution microbi - al ecology gains a new phase. This paper presents a mini-review on the molecular ecological techniques used for mierobial diversity research in contaminative environment based on 16S rDNA, as well as 16S rDNA sequence analysis, ARDRA (Amplitied rDNA Restriction Analysis), T-RFLP/RFLP (terminal restriction fragment length polymorphism), RISA ( ribosomal intergenic spacer analysis), SSCP(single strand conformational polymorphism),DGGE/TGGE (Denaturing/Temperature gradient gel eletrophoresis) and RAPD (Random Amplified Polymorphism DNA).
