RNA Interference and Its Related Application

摘要 RNA干扰(RNAi)现象是生物界的一种保守行为,能特异抑制同源基因的表达,具有抵抗病毒入侵和维持基因组稳定的作用,是生物体对病毒基因等外源核酸侵入的一种保护性反应。RNAi是由dsRNA诱导的多步骤反应,首先dsRNA被降解为21~25nt的siRNA,siRNA再与对应的mRNA特异结合、降解,从而阻止mRNA的翻译。当病毒在宿主细胞内进行复制、增殖时,病毒RNA可被降解成长19~23nt、具有5′磷酸和3′羟基及2个nt(dTdT或UU)3′外悬粘末端的siRNA,从而通过RNAi的作用抑制细胞内病毒的增殖。它的出现为生物学家提供了一种快速、简便的反向遗传学技术,在后基因组时代功能基因的研究中具有重要的应用前景,同时在人类基因治疗方面具有潜在的应用价值。
