摘要
△6-脂肪酸脱氢酶基因可将亚油酸转化为γ-亚麻酸。我们将克隆的深黄被孢霉△6-脂肪酸脱氢酶基因(D6D)插入植物表达载体pBI121,构建了重组质粒pBI121-D6DMI。通过农杆菌(Agrobacterium tumefaciens)介导途径,将深黄被孢霉△6-脂肪酸脱氢酶基因导入了甘蓝型油菜恢复系,经过诱导分化获得抗卡那霉素的再生植株。PCR检测及Souther杂交结果表明,外源基因△6-脂肪酸脱氢酶基因已整合到油菜再生植株的基因组中。
The desaturase of △^6-fatty acid (D6D) is a rate-limiting enzyme in the production of γ-linolenic acid(GLA) and it can converts linoleic acid(LA) and α-linolenic acid(ALA) into GLA. In this study, a △^6-fatty desaturase gone was cloned from Mortierella isabellina and the plasmid pTMICL6 was constructed by inserting D6D gene into plant high-efficient expression vector pBI121, This construct was used for Brassica napus transformation mediated by Agroubacterium tumefaciens LBA4404. Results from PCR and Southern blotting analysis with transformed plants indicated that the Mortierella isabellina ,△^6-fatty acid desaturase gene was integrated into Brassica napus genomes.
