摘要 目的探讨大鼠肌源性干细胞体外原代培养的方法,为临床应用自体肌源性干细胞注射治疗压力性尿失禁提供实验依据。方法采用胶原酶和胰蛋白酶等分离大鼠骨骼肌细胞,应用差速贴壁法进行纯化获取肌源性干细胞,在倒置显微镜下观察干细胞的形态,通过免疫组织化学法进行鉴定。结果通过差速贴壁法可获取高纯度的肌源性干细胞,免疫组织化学技术可鉴定其肌源性和干细胞特性。结论肌原性干细胞可通过体外原代培养获取,适用于组织工程和基因治疗,临床应用有待进一步研究。 Objective To establish the methods for culture and identification of rat skeletal muscle derived stem cells in vitro. Methods Rat MDSC were isolated from gatrocnemius muscle biopsies of normal 4-6 week-old-Sprague-Dawlay rats and purified by different adhesion time. CeLls that took approximately 5-6 days to adhere to the flasks were MDSC. The cells were observed by invert microscope and indentified with cellular immunochemieal stain. Results At least 90 % MDSC were harvest by the prelate technique and these cells were Sca- 1 + and desmin + by immunohistochemical staining. Conclusion It is easy to primary culture of MDSC in vitro and these cells can be used to tissue engineering and transgene therapy.
