摘要
为了研究可供人体应用的造血干细胞动员剂，将低相对分子质量葡聚糖硫酸酯（ＤＳ）给狗静脉注射，观察其对造血干细胞的动员作用。结果显示，注射ＤＳ１５ｍｇ／ｋｇ１ｈ后外周血中ＷＢＣ、单个核细胞（ＭＮＣ）和粒－巨细胞集落形成单位（ＣＦＵ－ＧＭ）计数开始升高，３ｈ达到峰值，每升血中的细胞计数分别为药前的１．６、２．１和４．５倍。外周血中ＷＢＣ、ＭＮＣ计数，随着ＤＳ剂量增加而增加，注射ＤＳ６０ｍｇ／ｋｇ时ＷＢＣ、ＭＮＣ数分别为药前的２．１和４．０倍。注射不同剂量ＤＳ〈以１５ｍｇ／ｋｇ或３０ｍｇ／ｋｇ组动物外周血中各系祖细胞升高幅度最大，ＣＦＵ－ＧＭ、ＣＦＵ－Ｍｅｇ、红系爆增式集落形成单位（ＢＦＵ－Ｅ）和ＣＦＵ－Ｍｉｘ每升血中计数分别为药前的９．０、７．２、１０．３和１３．
Healthy beagles about 1 3 years of age and weighing 7 11 kg were used.Dextran sulfate(DS) 500 mg/5 ml ampoule was injected.At 1 h after iv injection of DS,the numbers of WBC,MNC and CFU GM in the peripheral bolld increased and reached peak values at 3 h after injection,which were 1.6,2.1 and 4.5 folds of preinjection values respectively.They returned to normal levels at 7 h after injection.The number of WBC,MNC in dog bolld began to increase after injection of DS 7.5 mg/kg body weight.The greater the doses of DS,the higher the concentration of WBC and MNC in dog blood.15 30 mg/kg caused the best mobilization of CFU GM,CFU Mge,BFU E and CFU Mix/L blood,being 9.0,7.2,10.3 and 13.0 folds of preinjection values respectively.
