摘要
运用杂交瘤技术,制备出7株分泌抗人轮状病毒短型S_2株单克隆抗体的杂交瘤,其中6株分泌IgG_(2a),1株分泌IgG_(2b)抗体。7株杂交瘤培养上清的免疫荧光抗体(IFA)滴度为1:32~256,腹水IFA滴度为1:8000~25600。双夹心ELISA腹水抗体滴度达10^(-4)~10^(-6)。7株McAb均与HSV、CMV、RSV、ADV等其它常见病毒无交叉反应。7株McAb均能与HRV长型Wa株反应,滴度为10^(-3)~10^(-6)。将其中二株或数株McAb混合后用ELISA检测其相应的OD值较每一单株为高,但相加指数(AI)值均在10%左右。结果表明,7株McAb均属RV群特异性抗体,但各自作用的抗原决定簇可能稍有差异。
Using a strain of human rotavirus(HRV), S2 (Subgroupl)as an immunizing antigen, we prepared 7 clones of mouse-mouse hybridoma cell secreting monoclonal antibodies (McAbs)agalnst the immunogen itself. Six strains of Me Abs were identified as mouse IgG2a and 1 was IgG2b. The antibodies titres of culture fluids from 7 hybridoma cells were 1:32-256, and those of mouse ascites were 1:8000-25600 by indirect fluorescent antibody technics (IFA). By sandwich ELISA, the antibodies titres of mouse ascites were 10^-4-10^(-8). None of 7 McAbs reacted with other viruses including HSV,CMV, RSV and adenovirus. All 7 Mc Abs could react with HRV Wa strain, a long-type RV by PAGE ( Subgroup Ⅱ). Using ELISA addivity test, when 2 or more strains of Mc Abs were mixed, the maxium optical density (OD)value of the mixture was so- mewhat higher than that of single Mc Ab, but Addltivity Iadices (AI)of mixed and single McAbs were about 10% all. These results indicated that all 7 McAbs belonged to Group A of RV and the difference in the avidities of 7 MeAbs against HRV was caused probably by slight difference in the common antigenic determinant among various HRV strains.
