家蝇幼虫分泌物抗菌组分的抗菌效果观察及筛选 被引量:11

摘要 目的筛选家蝇幼虫分泌物中的抗蔺组分并测定其抗菌活性。方法用盐析、凝胶过滤层析、反相高效液相色谱(RP-HPLC)分离纯化分泌物中抗菌蛋白肽,以大肠埃希茼为指示曲种,采用平板滤纸片法检测抗菌活性,并观察其对金黄色葡萄球菌等多种细菌的抗菌活性。结果经50%~80%硫酸铵盐析沉淀、Sephadex G-100、G-50凝胶过滤往层析分离得到一活性峰,该活性峰埘大肠埃希菌及金黄色葡萄球菌等细菌显示较强的抗菌作用,进一步用反相高效液相色潜纯化,该活性峰由3个活性组分构成。结论家蝇幼虫分泌物中含有3个抗菌组分,对多种革兰氏阳性及阴性菌具有较强的抗菌活性。 Objective To isolate antibacterial peptides from Musca domestica larvae secretion and to study their antibacterial effect. Methods The antibacterial peptides were isolated and purified from secretion of Musca domestica larvae through ammonium sulfate dialysis, Sephadex chromatography and RP-HPLC. Their antibacterial activity was detected on agarose plates. Results After 50 80% ammonium sulfate pre cipitation and Sephadex G-100,G-50 filtration, an antibacterial fraction was obtained, which showed potent antibacterial activity on Escherichia coli and Staphyloccus aureus. Further purified by HPLC, the antibacte rial fraction was composed of three antibacterial peptides. Conclusion There were three antibacterial active ingredients in housefly larvae secretion that exhibited potent antibacterial activity to both Gram negative and positive bacteria.
