摘要 转录因子与DNA的相互作用是基因表达调控系统的重要组成部分。研究二者相互作用的特点,有助于阐明基因表达调控的规律。二者的相互作用取决于二者所特有的结构,以及在此基础上所产生的各种相互作用力。研究DNA-蛋白质复合物的方法主要分为2类:实验方法和生物信息学方法。目前已经有多种实验方法用于研究转录因子与DNA的相互作用,包括指数富集的配基系统进化技术、噬菌体展示技术及DNA微阵列技术等;以计算机为辅助的生物信息学方法也越来越广泛地用于二者相互作用的研究。 The interaction between transcription factors and their DNA recognition sites are important for the regulation of the network that controls gene expression. It is helpful to understand the regulatory pathways of gene expression by elucidating DNA-protein interactions. The interactions are controlled by their special structures. At present there are two kinds of methods available for the study of their interactions, including experimental methods and bioinformatic ones. There are several experimental methods such as systemic evolution of ligands by exponential enrichment (SELEX) , phage display (PD) , DNA microarrays, etc.. Bioinformatic techniques for predicting the interactions have led to considerable progress in recent years. This paper reviews the methods currently being employed in the study of protein-DNA interactions.
