应用免疫磁珠法分离人胚胎神经干细胞的培养及鉴定

摘要 目的:从流产胚胎中分离出神经干细胞并培养、扩增和进行鉴定,为神经干细胞移植寻找源泉。方法:实验于2001-02/2005-06在加拿大多伦多大学和南京大学附属鼓楼医院完成。收集流产的胎儿脑组织,备人胚胎大脑组织细胞悬液制免疫磁珠法分离神经干细胞,实验设计共随机选择24个分离的单个神经干细胞通过反转录酶聚合链反应技术的方法用DNA扩增仪检测单个细胞中nestin神经干细胞标记物,betaactin为内参对照。以结果:分离的人胚胎神经干细胞生长、扩增正常,nestin阳性细胞纯度极高。结论:免疫磁珠法对胚胎神经干细胞的分离,及反转录聚合酶链反应技术在神经干细胞鉴定中的应用是值得进一步探讨的途径。 AIM: To explore method of isolation, culture, amplification and identification of aborted embryonic neural stem cells in order to find new sources of transplantation of neural stem cells. METHODS: The experiment was conducted at the University of Toronto and Gulou Hospital affiliated to Nanjing University between February 2001 and June 2005, Brain tissues of aborted fetus were collected to make cell suspension of human embryonic cerebral tissue. Neural stem cells were isolated with immunomagnetic beads method, Nestin gene markers of 24 single cells chosen randomly were detected with reverse transcriptasepolymerase chain reaction (RT-PCR) with DNA amplification apparatus, and beta actin was considered as control, RESULTS: Growth and amplification of separated human embryonic neural stem cells were normal, Nestin gene markers were positive in most cells. CONCLUSION: Isolation of embryonic neural stem cells with immunomagnetic beads method and the application of RT-PCR in the evaluation of neural stem cells are worthy to be discussed further.
