香蕉果实β-半乳糖苷酶基因cDNA克隆及序列分析 被引量:10

cDNA Cloning and Sequencing of β-galactosidase Genes in Banana(Musa spp.) Fruit

摘要 利用已报道β-半乳糖苷酶基因(-βga lactos idase)的保守序列设计简并引物,进行RT-PCR,从香蕉果实中得到900 bp左右的一个片段,命名为M A-ga l,序列分析表明M A-ga l全长927 bp,编码309个氨基酸,具有所有β-半乳糖苷酶基因共有的活性催化部位GGP IILSQ IENEY(F);M A-ga l片段的氨基酸序列与芦笋、草莓、番茄、芒果及鳄梨等果实相应基因的相似性分别为85.8%、80.9%、80.5%、79.1%和76.3%. Degenerate primers were designed according reported conservative sequence of β-galactosidase genes and amplified by RT-PCR so that a fragment of about 900 bp were obtained;The fragment was named MA-gal and its sequencing indicated that MA-gal was 927 bp in its full length,encoded 309 amino acids and contained GGPIILSQIENEY(F) motif,the presumptive catalytic domain which was shared by all β-galactosidase genes;The amino acid sequence of MA-gal is similar to those of the corresponding gene fragments of asparagus ,strawberry, tomato ,mango and avocado with their homologies standing at 85.8 %, 80.9% ,80.5%,79.0% and 76.3% ,respectively.
