摘要
目的对透明质酸钠(HA)的结构进行表征。方法用红外光谱法测定HA的一级结构,采用拉曼光谱法作为佐证。在认识HA中存在的化学基因后,用圆二色谱测定HA的三级结构进行探索。结果在此实验条件下所得到的红外光谱与美国材料实验协会提出的谱图基本上吻合,同时出现新的吸收峰。拉曼光谱的吸收峰位置与红外光谱相一致,并且观测到低于500 cm-1的波数有吸收峰出现。HA的圆二色谱图在210 nm有特征性的负峰出现。结论HA分子结构中存在醇类羟基,并且羟基之间发生了很大程度的缔合,圆二色谱图中的负峰表示HA在溶液中的形态为β构象,这些是实现HA黏弹性的基础。
Purpose To characterize the adopted to measure the first structure of HA, knowledge of the groups in HA, circular structure of hyaluronan (HA). Methods Infrared spectrum was with the results of Raman spectrum as a proof. On the basis of dichroism was taken to explore the tertiary structure of HA. Results The Infrared spectrum acquired t, nder the condition of this essay mostly accords with that issued by ASTM, with some minor difference. The absorption peaks in the Raman spectrum of HA correspond to those in the Infrared spectrum, and some peaks may be observed below 500 cm^-1. A characteristic negative peak appears in the circular dichroism of HA. Conclusion Mellow hydroxyls exist in the hyaluronan molecule, with a large degree of association, and hyaluronan takes the form of β-folding in solution, which contributes to the viscosity of HA.
