摘要
目的:了解细胞信号转导过程中,内化作用如何调节HGF-cM et的信号转导,Dynam in蛋白对HGF-cM et内化作用中的影响。方法:以标记的HGF刺激体外培养的HepG2细胞,并在一组细胞中转染无功能的突变Dynam in蛋白,进行细胞摄入的对照实验。结果:突变Dynam in蛋白在37℃时降低了HGF-cM et的内化率约63%,而4℃时内化量一直维持较低水平。结论:HGF所涉及的信号转导由其受体cM et蛋白所介导,与细胞的内吞过程紧密相连,Dynam in蛋白在内化过程中起重要作用。
Objective:To investigate the internalization how to regulate the signal transduction of HGF-cMet, specially the influence of Dynamin on internalization of HGF-cMet by receptor inducing. Methods:Hepatocyte growth factor was labeled with ^125Ⅰ. The quantity of HGF internalization of ^125Ⅰ HGF in HepG2 cell at 4℃ and 37℃ and different time point was compared respectively with cell uptake assay. Results: Internalization of ^125Ⅰ-HGF in HepG2 cell was reduced about 63% in cell line of K44A mutation Dynamin transfected at 37℃. At 4℃, the level of interlization maintained very low. Concluslon:The process of signal transduction was induced by HGF receptor cMET and was related with internalization of cell. The danamin is important role in process of internalization.
