“双固一通”针法对帕金森病模型大鼠黑质多巴胺能神经元保护作用的实验研究 被引量:2

摘要 目的探询“双固一通”针法对帕金森病模型大鼠黑质多巴胺能神经元的保护作用。方法将Wistar大鼠40只,随机分为正常对照组、假手术对照组、模型组、双固一通组等4组,每组10只。采用6-OHDA单侧纹状体立体定向微量注射法制备大鼠旋转帕金森病模型。双固一通组在造模成功后取关元、足三里、风府、太冲四穴行电针治疗2周。然后,取各组动物中脑,采用流式细胞技术检测各组大鼠黑质多巴胺能神经元数量和细胞凋亡率。结果双固一通组毁损侧黑质TH阳性细胞百分比显著高于模型组(P<0.01),黑质细胞凋亡率显著低于模型组(P<0.01)。结论双固一通针法能提高帕金森病模型大鼠黑质多巴胺能神经元数量,降低凋亡率,对多巴胺能神经元具有明显保护作用。 To discuss the protective effect on dopaminergic neurons in substantia nigra of the parkinsonian rats by the acupuncture therapy of "Shuanggu Yitong". Methods:The parkinsonian rats were modeled by stereotaxic microquantitative injection in the right striatum with 6-hydroxydopamine and treated by the acupuncture therapy of "Shuanggu Yitotag". All the experimental rats were divided into four groups:normal group, sham-modeling group, model group and "Shuanggu Yitong"group. The"Shuanggu Yitong"group was treated by needling the four acupoints of"Fengfu","Taichong","Guanyuan"and"Zusanli"dter modeling. After the treatment of 2 weeles,the therapy of flow cytometry is adopted to detect the percentage of the dopaminergic neurons and apoptotic cells in the substantia nigra. Results: The percentage of TH positive neurons in the destructive side of the suhstantia nigra of the"Shuanggu Yitong" group is obviously higher than thatof the model group(P〈0. 01),simultaneously the percentage of apoptosis of substantia nigra of the destructive side of the needling group isobviously lower than that in model group (P〈0. 01). Conclusion. The needling therapy of "Shuanggu Yitong"demonstrate the protectiveeffect on dopaminergic neurons in substantia nigra of the parkinsonian rats.
