布洛芬对Aβ_(25-35)所致大鼠海马损伤及iNOS表达的影响

摘要 目的研究非甾体类抗炎药布洛芬(Ibuprofen,Ibu)对大鼠脑室内注射Aβ25-35所致海马损伤及iNOSmRNA表达的影响。方法大鼠灌胃给予Ibu(7.5,15mg.kg-1)连续应用3w,脑室内注射Aβ25-35(5μL,2M),注射后1w,取脑组织,进行尼氏(Nissl)染色和胶质纤维酸蛋白(glial fibrillary acidic protein,GFAP)免疫组织化学染色,观察海马CA1区锥体神经元形态学改变和星形胶质细胞的活化浸润情况。RT-PCR分析诱导型一氧化氮合酶(iNOS)基因表达水平。结果大鼠脑室内注射Aβ25-35可引起海马CA1区星形胶质细胞的激活和浸润及锥体神经元的损伤,同时伴有iNOS mRNA表达增加。Ibu(7.5,15mg.kg-1,连续应用4w)能减轻海马CA1区锥体神经元的损伤及星形胶质细胞的激活和浸润,抑制Aβ25-35引起的iNOS mRNA表达增加。结论Ibu能够通过抑制Aβ25-35引起的iNOS mRNA表达,减轻大鼠海马锥体神经元的损伤。 Objective To study the Effects of ibuprofen on Aβ25-35-induced hippocampal damages and iNOS expression in rats. Methods Rats were given ibuprofen (7.5 , 15 mg·kg^-1 daily, ig) for 3 weeks prior to icy, Rats were injected icy with Aβ25-35 (5μL, 2M) . 1 week after injection, Nissl staining and GFAP immunohistochemical technique were employed to determine the morphology of pyr-amidal neurons and astrocyte infiltration in hippocampal CA1. The expression of iNOS mRNA was determined by Reverse transcription- PCR analysis. Results Intracerebroventricular injection of Aβ25-35 in Rats elicited astrocyte activation and infiltration and the loss of pyramidal neurons in hippocampal CA1, which was accompanied by the increase of iNOSmRNA expression in hippocampal CA1. Ibuprofen (7.5, 15mg·kg^-1) significantly reduced the Aβ-induced astrocyte activation and infiltration , the loss of pyramidal neurons in hippocampal , and reversed the Aβ25-35- induced increases in iNOS mRNA expression. Conclusions Ibuprofen attenuates damages of pyramidal neurons in hippocampal CA1 by decreasing the Aβ25-35 - induced iNOS mRNA expression.
