Role of low estrogen in follicle/oocyte maturation and embryo development

摘要 目的探讨雌激素在卵细胞和胚胎发育中的作用。方法采用3、4周C57B6杂交1代小鼠,实验组小鼠每日腹腔注射芳香化酶抑制剂arimidex100μg,对照组注射等体积的生理盐水。第2天腹腔注射7.5Iu妊马促性腺激素诱导卵泡发育。2d后测定雌、孕激素水平并观察卵巢形态;注射妊马促性腺激素47h后,腹腔注射5.0Iu绒毛膜促性腺激素,两只雌鼠与一只雄鼠同笼饲养,2d后测定雌、孕激素水平并取出胚胎体外培养,连续3d观察胚胎发育情况。结果实验组的雌激素94%被抑制,孕激素75%被抑制,两组小鼠的平均卵泡数差异无显著性(对照组平均每个卵巢有30.4个卵泡,arimidex组有27个卵泡)。实验组的雌激素水平较对照组明显降低(P<0.01),孕激素水平降低不明显(P>0.05)。两组的平均胚胎数、体外培养桑椹胚生成率、囊胚生成率、孵化囊胚生成率差异无显著性(P=0.2,0.1,0.44,0.38)。结论雌激素水平的升高对卵泡的成熟和胚胎本身的发育无决定作用。 [Objective] To study the requirement of estrogen for follicular and embryo development. [Methods] Synthesis of estrogen was blocked by injection of aromatase inhibitor arimidex 100ug/mouse to 3, 4 weeks old C57B6 F1 female mice each day, the control group were given the same amount saline. Exogenous gonadotrophin (7.5 Iu pregnant mare serum gonadotroohin, PMSG) was administered to induce follicular growth and development the second day. Experiment one we tested estrogen and progesterone and examined ovary morphology two days later. Experiment two 47 hours after PMSG, 5.0 Iu hCG was given and two female mice caged with a male mouse. Two days later we tested estrogen and progesterone and took out embryos and cultured them then examined the embryo development every 24 hours for 3 days. [Results] Before hCG injection, the cstradiol level was suppressed by 94%, the progesterone level was suppressed by 75%, there was no difference in the mean number of total follicles found in each animal of either groups (30.4 follicles in the control and 27 in the arimidex group). 2 days after hCG injection, the estradiol level of arimidex group was significantly lower comparing with control group (P〈0.01), the progesterone level of arimidex group was not obviously lower ( P 〉0.05). The embryos, morula, blastocyst and hatching blastocyst from two groups were not signifieandy difference (P=0.2, 0.1, 0.44, 0.38 respectively). [Conclusions] In the present study, by deprving mice of the normal E support, we have been able to rule out the absolute need for rising levels of E for completion of the follicular maturation process and development of embryos in vitro.
