全反式维甲酸对HL-60细胞NF-kB、CyclinD1表达的影响 被引量:4

摘要 目的探讨全反式维甲酸(ATRA)对HL-60细胞周期、NF-kB和Cyclin D1表达的影响。方法采用不同浓度的ATRA作用于HL-60细胞,于48h后收集细胞,用流式细胞术分析HL-60细胞周期、NF-kB和Cyclin D1表达的变化。结果维甲酸组HL-60细胞G1期细胞比值升高,S期细胞比例下降,NF-kB、Cyclin D1表达均下降,其中10-7M、10-6M浓度组与对照组相比差异均有显著性(P<0.05),而10-8M浓度组却差异均无显著性(P>0.05);维甲酸10-6M浓度组NF-kB、CyclinD1的表达呈显著正相关(r=0.55,P<0.01,n=12)。结论全反式维甲酸(ATRA)通过下调NF-κB抑制CyclinD1活性表达,可能是其阻滞细胞周期诱导HL-60细胞分化的机制。 【Objective】 Study the effects of ATRA on cell cycle and expression of CyclinD1 and NF-kB. 【Methods】 HL-60 cells were cultured in RPMI 1640 medium with NS and ATRA in the final concentrations of 10^-8 M, 10^-7 M and 10^-6 M, respectively. After culturing for 48 h, the expression of NF-kB, cells cycle and CyelinD1 were examined by FCM. 【Results】 Ratio of G1 cyelc cells in 10^-7 M and 10^-6 M groups significantly increased than those in control group (P〈0.05), while there is no significance in 10^-8 M group (P〉0.05), Ratio of S cycle cells in 10^-7 M and 10^-6 M groups significantly decreased than control group (P〈0.05). The expression of NF-kB and CyclinD1 in 10^-7 M and 10^-6 M groups significantly decreased than those in eontml group (P〈0.05). There was a good positive link between the expression of NF-kB and CyelinD1 in 10^- M groups (r=0.55, P〈0.01, n=12). 【Conclusions】 All-trams retinoic acid could down-regulate CyelinD1 expression by restraining expression of NF-kB, then induce differentiation of HL-60 cells.
