摘要
目的探讨尼美舒利对体外培养的结肠癌细胞生长及细胞周期分布的影响。方法以人结肠癌细胞株HT-29、HCT-116为对象,用体外药物敏感实验(MTT)法检测尼美舒利对肿瘤细胞的增殖抑制效应,流式细胞术检测肿瘤细胞周期分布变化情况。结果(1)尼美舒利对人结肠癌细胞株生长的抑制作用呈时间、剂量依赖性效应,且对HT-29细胞作用强于HCT-116细胞;(2)尼美舒利可改变结肠癌细胞株细胞周期的分布,明显降低增殖指数。结论尼美舒利可通过抑制COX-2酶活性而抑制肿瘤细胞的分裂和增殖,诱导其凋亡,干预结肠癌的发展。
Objective To evaluate the potential role of nimesulide on carcinoma cell lines, Methods The human colon cancer cell lines HT 29 the proliferation and cell cycle distribution of colon adenoand HCT-116 were employed in the study. After treatment with nimesulide, the inhibitory effect of nimesulide on the proliferation of cancer cells was quantified by MTT assay, and the cell cycle distribution of tumor cells was detected by flow cytometry. Results (1) Nimesulide inhibited the proliferation of the tumor cells in time and dose-dependent manners. The inhibitory effect on HT-29 cells was stronger than that on HCT-116 cells. (2)Nimesulide changed cell cycle distribution of both cells; the proliferation index of cells was obviously decreased also. Conclusion Nimesulide may inhibit the division and proliferation of cancer cells through the inhibition of COX-2 activity, resulting in the apoptosis of tumor cells and interfering in metastasis and relapse of colon cancer.
