摘要
本文叙述应用自行研制的激光微探针飞行时间质谱仪及采用近年发展起来的基体辅助激光解吸的新方法对多种已知蛋白质的分子量进行测量,表明此法比一般的常规方法具有速度快(10分钟1个样品)、准确度高(±1%~0.1%)、灵敏度高(10-12~10-15mol)等优点。同时我们结合SDS-PAGE等生化方法,对蛋白质、多肽、伴花生蛋白酶和合成多聚物的分子量进行测定,也取得了令人满意的结果。
Matrix-assisted laser desorption mass spectrometry(MALDI-MS) has been used to determine the molecular weight of proteins with a home-made laser microprobe time offlight mass spectrometer.It demonstrated that this new method is possesed of high accuracy(±1 % ̄ 0.1%),high sensitivity(10-12 ̄10-15mol) and rapidity(with few minutes),which are far better than those traditional methods.By the combination of SDS-PAGE and degradation,we had determined the molecular weight of proteins,polypeptides,conarschin protein and synthesized polymers with satisfied results.
