摘要
通过PCR方法自含鸡白细胞介素18(Chicken interleukin-18,ChIL-18)成熟蛋白(mature ChIL-18,mChIL-18)基因的克隆质粒中扩增mChIL-18基因,并将其克隆到真核表达载体pcDNA3.1/V5-His-TOPO中。阳性克隆鉴定后,在脂质体作用下转染鸡胚成纤维细胞(CEF),通过间接免疫荧光试验(IFA)检测到了mChIL-18在CEF中的表达。
Chicken interleukin 18 mature protein gene was amplified from the recombinant plasmid named pT-mChIL18 by polymerase chain reaction(PCR) and cloned into pcDNA3.1/V5 His TO PO. The recombinant plasmid was transfected into chicken embryonic fibroblasts(CEF) with li posome. 24,48,72 hours after transfection, the expression of mChIL-18 were successfully tested by indirect immunofluorescence assay(IFA) with the antiserum of mChIL-18 expressed in E. coli.
