摘要
目的 观察以Veto细胞大规模生产狂犬病毒时的细胞生长和病毒感染特性。方法 以分种和混种方法,经过不同培养时间,测定Vem细胞的生长状况和胞内外病毒含量。结果 细胞形态随接种病毒时间的长短有所变化,不同接种方法在病毒滴度也无显著性差异。培养上清夜病毒含量以第4~9d滴度最高,细胞病毒感染率在第4d达80%以上,第10d后开始下降,第15d降入低谷。结论 以0.01~0.10MOI的加毒比例,培养第4,9d收获效果最佳。
Objective To explore the growth and virus infection characteristics of Veto cell producing rabies virus in great quantity. Methods The growth status of Vero cell and control of virus inside and outside after cultured for various durations by separate inoculation and re - inoculation. Results The re is certain change in morphology of Vero cell along with the progress of time after inoculation and no significant differences were observed in virus titer with different inoculation methods. The content of virus in spernatant was the highest during the fourth and ninth day, while the infection rate of virus was over 80% on the fourth day, and began to drop on the tenth day, and reduced to he lowest on the fifth day. Conclusion The proportion virus be at the range of 0.01 ~0.10 MOI and the optimum recovery time be during the fourth and ninth day after culture.
