计算液相色谱死体积的新方法

A New Method of Calculation the Dead Volume in HPLC

摘要 本文测定了苯的同系物在甲醇、乙醇、丙醇、乙腈和四氢呋喃分别与水组成两元流动相中,不同配比和不同柱温在ODS柱上的保留体积,用反磁化率的二次曲线法求取反相液相色谱的死体积。用同系物的校正保留值与碳数间的线性关系进行校验,相关系数接近于1。结果表明死体积与流动相种类、配比和柱温等因素有关。 The retention volume of benzene series in binary mobile phase composed of different compositions of methanol, ethanol, propanol, acetonitrile, THF with water respectively, in the ODS column with different temperature, were studied. The dead volumes in RPLC were measured by the method of antima-gnetic quadratic curve. The linear relationship between the retention volumes of the series and the carbon numbers wes verified and the correlation coefficients obtained were nearly equal to 1. Experimental results further showed that dead volumes were also related to factors, such as the characteristics of the mobile phase, its composition and the column temperature.
