摘要
对白桦花序进行分期取材,以CTAB法提取各期材料的总RNA,应用SMART(Switching Mechanism At5′end of RNA Transcript)策略合成cDNA。根据BpHEN序列片段设计引物,通过特异扩增、产物纯化、梯度稀释,制备出系列标准样。通过实时定量PCR分析BpHEN在各取材时期的表达情况表明,BpHEN在雌花序中的表达要高于雄花序,且在雌花序中有3个表达高峰,分别对应于授粉期、胚珠发育期、果实发育期。
Betula platyphylla inflorescence was sampled in floral phase with temporal interval. Total RNA was isolated from the samples of every period by the method of CTAB, which was subsequently used to synthesize cDNA based on Switching Mechanism At 5'end of RNA Transcript (SMART) strategy. Primers were designed according to the sequence of BpHEN, a gene related to floral development, and then standard samples were prepared after specific amplification, purification, and dilution with series of grads. Exprcssion of BpHEN was analyzed by the method of real-time PCR. Result showed that BpHEN was more highly expressed in female inflorescence tban in male inflorescence, and three peaks corresponding to pe- riods of pollination, ovule development, fruit development were found in female inflorescence.
