实验性阻塞性肺气肿大鼠的凝血-纤溶系统失衡 被引量:3

摘要 目的:观察实验性阻塞性肺气肿大鼠的凝血纤溶系统失衡状态,并对其机制进行探讨。方法:Wistar大鼠24只,随机分为2组,正常对照组和模型组,每组12只。采用熏香烟加尾静脉注射内毒素的方法制备实验性阻塞性肺气肿大鼠模型。实验结束后,观察各组大鼠的病理及肺功能情况,用酶联免疫吸附法(ELISA)测定血浆组织型纤溶酶原激活剂(tPA)、纤溶酶原激活物抑制剂-1(PAI-1)、组织因子途径抑制物抗原(TFPI:Ag)的含量,发色底物法测定血浆抗凝血酶-Ⅲ(AT-Ⅲ)活性。结果:模型组大鼠肺功能指标第0·2s用力呼出容积占用力肺活量百分比(FEV0·2/FVC)(68·25±8·90)%,与正常对照组(88·52±2·09)%比较,差异有显著性(P<0·01);模型组大鼠AT-Ⅲ和TFPI分别为(56·04±14·81)%、(1·39±0·26)ng/mL,与正常对照组(85·46±18·03)%、(1·05±0·17)ng/mL比较,差异有显著性(P均<0·01);模型组大鼠tPA和PAI-1分别为(2·1±0·73)ng/mL、(0·54±0·07)ng/mL,与正常对照组(1·45±0·43)ng/mL、(0·80±0·24)ng/mL比较,差异有显著性(P均<0·05)。结论:模型组大鼠体内的凝血纤溶系统存在失衡状态,AT-Ⅲ、TFPI、tPA和PAI-1可能参与这一过程。 Objective:To observe the abnormalities of eoagulatin and fibrinolytic system in experimental pulmonary emphysema in rat. Method:24 Wistar rats were randomly divided into two guoups, the normal group(A) and the model group(B). Rats in group B were injected endotoxin by the vena caudalis and exposed to the smoke. Pumonary function indices and the histopathological changes were evaluated. The blood plasma levels of tPA, PAI-1 and TFPI: Ag were measured by ELISA method, the blood AT+ Ⅲ activity were measured by Chromogenic assay. Result: Compared with group A, group B presented significant differences in FEV0.2/FVC (68.25 ± 8.90)% vs (88.52 ± 2.09)%, (P 〈 0.01 ); the plasma levels of AT-Ⅲ activity, TFPI: Ag, tPA and PAI-1 [ (56.04 ± 14.81)% vs(85.46±18.03)% , (1.39±0.26)ng/mL vs(1.05±0.17)ng/mL, (P〈0.01);(2.1 ±0.73)ng/mL vs ( 1.45± 0.43) ng/mL, (0.54 ± 0.07) ng/mL vs (0.80 ± 0.24) ng/mL ( P 〈 0.05)]. Conclusion: There were abnormalities of coagulatin and fibrinolytic system in experimental pulmonary emphysema in rat and the AT- Ⅲ, TFPI, tPA and PAI-1 might be involved in the process.
