摘要
目的了解电阻抗法检测全血血小板聚集(ADP为诱导剂)试验的影响因素。方法用Chrono-log560型血小板聚集仪的电阻抗通道检测10例健康体检者全血血小板的聚集(ADP为诱导剂)。每人抽血11.4ml,分别用肝素、枸橼酸钠抗凝,平均分为6管(各2ml),在不同的孵育温度(室温、37℃、4℃)与不同的放置时间(0min、30min、60min、120min)下检测血小板聚集情况(电阻值表示)。结果肝素与枸橼酸钠两种抗凝剂结果分别为10.35±5.98Ω,7.30±3.41Ω,二者有显著性差异(P<0.01);肝素抗凝血在室温、37℃时,血小板的聚集随时间的变化先升高后降低,但没有显著性差异(P>0.05),在4℃时,血小板的聚集随时间的变化而降低(P<0.05);枸橼酸钠抗凝血在不同的孵育温度及时间下,血小板聚集的变化不明显(P>0.05)。结论枸橼酸钠抗凝血适合于临床全血血小板聚集的检测,但需考虑红细胞压积对其的影响。
Objective: To investigate the effect factors of platelet aggregation in whole blood determinated by impedance aggregometer. Methods: 10 healthy people,s platelet aggregations in whole blood were determinated by Chrono-log 560 type impedance aggregometer with ADP inducer. To each person, 11.4ml venous blood was drawn and equally divided into 6 tubes, 3 of which were anticoagulated with heparin, the other 3 with sodium citrate. And pIateIet aggregation in whole blood placed at 3 different temperatures (room temperature ; 37℃ ; 4℃ ) and 4 different times (0min ; 30min ; 60rain ; 120min) was detected . Reasults. The results of the anticoagulants heparin and sodium citrate were 10. 35±5. 98Ω and 7. 30± 3. 41Ω respectively, having significant difference (P≤0. 01). Platelet aggregability in the venous blood anticoagulated by heparin first increases then decreases as time elapses at room temperature and 37℃ after venipuncture, having no significant difference (P 〉0. 05). Platelet aggregability decreases as time elapses at 4℃ (P 〈 0. 05). Platelet aggregability in the venous blood anticoagulated by sodium citrate doesn,t vary obviously at different temperatures and different times after venipuncture (P 〉 0. 05). Conclusions: In the clinical setting,the venous blood anticoagulated by sodium citrate is more suitable for platelet aggregation test in whole blood , but the effect of HCT must be considered.
