摘要
目的:研究选择性磷酸二酯酶4抑制剂(咯利普兰)对哮喘小鼠肺部树突状细胞(DC)数量的影响。方法:BALB/c小鼠24只随机分为3组,即对照组、哮喘组、咯利普兰组。采用免疫组织化学染色技术检测小鼠肺、气道树突状细胞;计算机图像分析技术对树突状细胞进行定量测定。结果:对照组小鼠的气管、支气管、肺泡和脏层胸膜构成了一个完整的NLDC-145+DC网络,对照组肺内NLDC-145+DC数量为(60±14.6)个/mm2上皮面积;哮喘组小鼠肺内NLDC-145+DC数量(210±35.6)个/mm2,较对照组显著增加(P<0.01);哮喘小鼠经咯利普兰处理后,肺内DC数量明显低于未处理的哮喘组犤(88±24.3)个/mm2vs(210±35.6)个/mm2犦。结论:树突状细胞参与了哮喘的气道炎症过程;咯利普兰可以下调肺哮喘小鼠肺部DC的数量,对哮喘的治疗具有潜在的应用前景。
Objective To investigate the effect of ehctive phosphodiesterase 4 (PDE4) inhibitor(mlipram) on the number of pulmonary dendritic cells(DC) in murine models. Methods Twenty-four BALB/c mice were randomly divided into three groups: control group, asthmatic group and rolipram-treatment group. The animal models of asthma was established by ovoalbumen (OVA) sensitizing-challenging,24 h after the last challenge. The number of DC in the pulmonary and tracheal samphs were performed with immunohistochemical methods and computer image analysis quantitatively. Results After OVA challenge, the whole amount of dendritic cells in tracheal, bronchus and alveolar were revealed by horseradish peroxidase staining using the NLDC-145 monoclonal antibody. The number of NLDC-145^+DC in the bronchial mucosa of asthmatic mouse was more than that in control group (P 〈 0.01). After seven days treatment with rolipram,the number of NLDC-145^+DC was less than that in asthmatic group (P〈 0.01). Conclusion It is suggested that dendritic cells are involved in the inflammation of airway of asthmatic murine and that rolipram may down-regulate the number of dendritic cells.
