摘要
目的观察大鼠去卵巢后体温变化与下丘脑视前区5-羟色胺1A(5-HT1A)受体亚型表达之间的关系。方法用数字温度计TM902C检测大鼠的肛温,酶联免疫吸附试验测定血清激素水平,根据5-HT1A受体互补DNA序列合成相应的特异性引物,用聚合酶链反应(PCR)法观察大鼠去卵巢后下丘脑视前区5-HT1A受体表达。结果 (1)大鼠去卵巢(OVX) 组自第8周末肛温明显高于假去势(sham)组(P<0.05)。(2)OVX组大鼠血清17β-雌二醇(17β-E2)较sham组下降,黄体生成素(LH)水平升高,差异显著(P<0.05)。(3)OVX组下丘脑视前区5-HT1A受体表达增强,与sham组比较有显著性差异(P <0.05)。结论大鼠卵巢摘除后体温升高,下丘脑视前区5-HT1A受体表达增强,提示5-HT在低雌激素状态下的体温调节中起一定作用。
Objective: To observe the relationship between the body temperature change and the expression of 5-HT1A receptor subtype in preoptic area of anterior hypothalamus of ovariectomized (OVX) rats. Methods: The rat rectal temperature (RT) was measured by the digital thermometer TM902C. Enzyme-linked immunoadsorbent assay (ELISA) was used to detect the serum hormone levels. The method of polymerase chain reaction (PCR) was used to detect the presence of 5-HT1A receptor subtype in the preoptic area of anterior hypothalamus of ovariectomized rat. The specific oligonucleotide primers were synthesized based on the complementary DNA sequences for the rat 5-HT1A receptor subtype. Results: (1) The RT in OVX rats was significantly elevated from the 8th wk after ovariectomy compared with that of sham-operated controls (P〈0.05). (2) Compared with the control group, concentrations of serum E2 decreased in the OVX group (P〈0.05), while concentrations of serum LH was increased(P〈0.05). (3) The expression of 5-HT1A receptor subtype in preoptic area of anterior hypothalamus of the OVX rat was significantly higher than that of control group. Conclusion. The RT was elevated after ovariectomy and the expression of 5-HT1A receptor subtype in preoptic area of anterior hypothalamus of the OVX rat was significantly higher, which suggest that serotonin may play some roles in thermoregulation in the state of low hormone concentrations.
