摘要 目的观测δ阿片受体激活对培养的心肌细胞是否有直接的促生长作用.方法体外培养乳大鼠心肌细胞.结晶紫法和[3H]TdR法测定心肌细胞的增生;流式细胞仪测定细胞周期百分比;Lowry等法测定培养细胞蛋白质含量;倒置显微镜下计数心肌细胞搏动频率.结果δ阿片受体激动剂D-丙(2)-D-亮(5)脑啡肽(DADLE)10 nmol·L-1~10 μmol·L-1能浓度依赖性促进细胞增殖,增加细胞蛋白质含量, 增加细胞搏动频率;高选择性δ阿片受体拮抗剂纳曲吲哚(naltrindole) 10 μmol·L-1对DADLE的促进作用有一定的抑制作用.结论δ阿片受体激活对体外培养的心肌细胞生长有促进作用. AIM To study if δ-0pioid receptor activation has direct effect on cultured cardiomyocyte growth. METHODS Myocardial cells of neonatal rats were cultured in vitro. The cellular proliferation was determined by crystal violet uptake and [^3H] TdR incorporation; the percentage of S + G2 + M in cell cycle was determined by flow cytometry; the protein content was assayed with Lowry, et al method ;contracting frequency of cultured myocardial cells was counted under the inverted microscope. RESULTS A 3-0pioid receptor agonist [ D-Ala^2, D-Leu^5 ]-enkephalin (DADLE) at 10 rlmol·L^- 1 _ 10μmol·L6- 1 concentration-dependently promoted the myocardial cells proliferation, increased protein content in culture cells and contracting frequency of the myocardial cells. Highly selective δ-opioid receptor antagonist naltrindole at 10 μmol·L^-1 inhibited the promoting effects of DADLE. CONCLUSION δ-Opioid receptor activation can promote growth in cultured myocardial cells.
