摘要 膜蛋白在细胞中执行着一些非常重要的功能。膜蛋白的溶解性是膜蛋白质组分析面临的主要挑战。膜蛋白溶解性的提高,使得经典的基于凝胶的蛋白质组策略和新兴的鸟枪蛋白质组策略在膜蛋白质组分析中逐渐被广泛应用,进而可以揭示膜蛋白的拓扑学特征和鉴定其翻译后修饰,为深入研究其重要的生物学功能积累数据。 Membrane proteins carry out many essential celluar functions. The formidable challenge in proteomic analysis of membrane proteins is their low solubility, effective enhancement of which makes gradually extensive application of classically gel-based and the latest shotgun proteomic strategies to membrane proteomic analysis. Furthermore, the characterization of membrane protein topology and its post-translational modification through these analyses can provide indispensable insights into biological functions of membrane proteins.
