摘要
目的观察氟尿嘧啶联合低分子肝素兔眼内灌注对视网膜组织的毒性损伤,探讨临床使用的安全性。方法健康纯种新西兰白兔24只,随机分为两组:A组和B组各12只。均取左眼为实验眼,将低分子肝素和氟尿嘧啶注入到BSS中,配成浓度为5 IU/mL的低分子肝素、0.25mg/mL氟尿嘧啶的混合灌注液。进行玻璃体切割手术的同时进行此药物灌注,药物灌注时间A组为30min,B组为60min。术后进行裂隙灯显微镜和间接检眼镜检查,2周时进行视网膜组织学检查。结果两组术中及术后观察过程中玻璃体腔均无明显出血。电镜检查:A组无异常发现;B组切片中可见到神经纤维层肿胀,色素上皮层细胞空泡变性等改变。结论兔眼对浓度为5 IU/mL的低分子肝素、0.25mg/mL的氟尿嘧啶的灌注液有较好的耐受性。
Objective To evaluate the ocular toxicity of combined application of 5-fluorouracil (5-fu) and low molecular weight heparin (LMWH) in rabbit eyes. Methods Twenty-four New Zealand rabbits were divided into two groups-30 min groups and 60 min groups. The infusion fluid contained 5-Fu and LMWH. Concentration is 0.25 mg/mL and 5 IU/mL respectively. After operation, a slit-lamp biomicroscope and indirect ophthalmoscope were used to examine every day. The tissue of enucleated eye in vitreous groups on the 14th day were examined by light microscope and transmission electron microscope. Results No evidence of haemorrhage was found in vitreous both 30 min and 60 min infusiongroups. In 60 min infusion occurred RPE cell and some nerve fibre layer denaturalization in retina. Histologic studies showed no toxicity in 30 min group yet. Conclusion There is well tolerance on rabbit eye to 5-FU and LMWH.
