摘要
介绍一种特异、灵敏、简便的蛋鸡血清载脂蛋白AI(apoAI)浓度单向免疫扩散测定法。用磷钨酸镁反复沉淀制备得高密度脂蛋白,经纯化、B radford法蛋白定量后(3.208 mg/mL),作为新西兰白兔的免疫原及载脂蛋白AI测定的标准。本室免疫所得兔抗鸡apoAI抗血清制成免疫琼脂板,加样培养进行单向免疫扩散测定。本法的标准曲线相关性良好,可信性、重复性满意。
This paper introduces a method for mensuration of apoAI from the serum of laying hens by SRID,which is specific, sensitive and convenient. ApoAI was separated from HDL which was isolated and purified from pooled serum by repeated precipitation with phosphotungstic acid - Mg. The concentration of it was mensurated by the method of Bradford （3. 208 mg/mL）. Then the apoAI was saved as immune original and first criterion of its mensuration . By immunizing New Zealand male rabbit for several times , the high specific antiserum was got. By using it, the immune agarose board was prepared, then the mensuration by SRID was carried out. The pertinency of this method＇s standard curve is good. Its creditability and repetition are satisfactory.
