摘要
目的探讨高强度聚焦超声(HIFU)终止早孕的机制。方法妊娠第8天SD大鼠48只,每组8只,用780KHz连续超声波5组超声声强开腹辐照子宫胚胎着床点各90秒,分别于妊娠第8天和15天取材,设妊娠第8天大鼠胚胎为对照组。应用免疫组化法检测早孕大鼠胚胎着床点蜕膜组织中白血病抑制因子(LIF)和基质金属蛋白酶9(MMP9)的表达。结果各辐照组LIF和MMP9早孕蜕膜组织中表达明显小于对照组(p<0.01),并随声强的增强而表达逐渐减弱。结论LIF、MMP9的变化可能是使HIFU终止大鼠早孕的关键。
Objective To explore the mechanism that High Intensity Focused Ultrasound (HIFU)terminates rat early pregnancy. Methods 8th-day pregnancy SD rats were randomly divided into different groups (each group number = 8). In experimental groups early pregnancy rats' implantation sides of uterus were irradiated by continuous ultrasonic wave of 780KHz with five different intensities of HIFU for 90 seconds. In experimental rats deciduas of implantation sides were obtained by 8th day and 15th day of pregnancy. The last group, without irradiating, was the 0W/cm^2 group. The expressions of leukemia inhibitory factor and Matrix Metalloproteinase-9 in all specimens were essayed by the immunohistochemistry. Results In experimental groups the expressions of LIF and MMP-9 in deciduas significantly decreased markedly in 0W/cm^2 groups (p〈0. 01). Conclusion The changes of LIF,MMP-9 may be the key mechanism of HIFU terminating rat early pregnancy.
