工频磁场对细胞缝隙连接通讯功能的影响 被引量:10

Effects of magnetic field on gap junctional intercellular communication

摘要 为了探讨极低频(ELF)磁场可能存有的促癌作用或协同促癌作用,观察了50Hz磁场对CHL细胞间缝隙连接通讯(GJIC)功能的影响。利用离子电渗注射法将荧光染料引入细胞内,以5分钟后的荧光偶合细胞(DCC)数作为GJIC功能的指标。研究了不同浓度的十四酰基咐拜醇酯(TPA)、不同强度的工频磁场及工频磁场与TPA(5ng/ml)共同作用对CHL细胞GJIC的影响。结果表明,TPA对GJIC的抑制具有浓度依赖关系,TPA抑制CHL细胞GJIC的阈浓度在1~5ng/ml之间;CHL细胞经0.80mT的工频磁场暴露24小时后的DCC数降至6.08±1.59,与空白对照组(9.84±2.27)比较,差异有显著性(P<0.05);0.20,0.40,0.80mT强度的工频磁场与5ng/mlTPA共同作用后的DCC数分别降至5.52±1.53,5.00±1.22,4.00±1.29,与单用5ng/mlTPA组(6.38±1.39)比较,差异也有显著性(P<0.05)。说明一定强度的工频磁场具有抑制或协同抑制GJIC的作用,提示其可能存有促癌或协同促癌作用。 In order to explore if the extremely low frequency(ELF)magnetic field can act as cancerpromoter or be synergistic with TPA in cancer promotion,the effect of 50 Hz magnetic field exposureand TPA on gap junctional intercellular communication(GJIC) of CHL cells was observed. Lucifer dyewas led into CHL cells by iontophoretic injection and the nurnber of dye-coupled cells(DCC)five min-utes after injection was adopted as the index of GJIC, The results showed that the suppression of TPAon GJIC was dependent on its concentrations and the threshold concentration of TPA was between 1~5ngm/ml;after exposure to 0.80 mT magnetic field for 24 hours,the number of DCC decreased to 6. 08±1. 59 whereas that in control group was 9.84±2.27(P<0. 05);when 5 ng/ml TPA treatrnent combinedwith exposure to 0.20,0.40 and0.80 mT magnetic field respectively,the number of DCC was 5.52±1. 53,5. 00±1. 22,and 4. 00±1. 29 respectively,whereas that in the group treated with 5 ng/ml TPAaIone was 6. 38± 1.39(P<0.05).It is suggested that certain intensities of 50 Hz magnetic field expo-sure might act as cancer promoter or be synergistic with other cancer promoters.
