肝素对人乳腺癌细胞生长及Hyase、HA和CEA表达的影响

摘要 目的探讨肝素是否具有肿瘤细胞毒性作用以及对肿瘤侵袭、转移相关因子如透明质酸酶(hyaluronidase,Hyase)、透明质酸(hyaluronic acid,HA)和癌胚抗原(CEA)的表达有何影响。方法用MTT法测定肝素对人乳腺癌细胞株MDA-MB-231生长的影响;酶联吸附法测定8株人乳腺癌细胞培养上清液中Hyase、HA和CEA的表达。结果肝素对人乳腺癌细胞MDA-MB-231的生长无显著影响,但能降低人乳腺癌细胞Hyase的表达(P<0.05),对HA和CEA的表达无显著影响(P>0.05)。结论肝素是一种非细胞毒性药物,不能直接杀灭肿瘤细胞,但能降低肿瘤侵袭、转移相关因子Hyase的表达。 Objective To evaluate the effects of heparin on tumor toxicity and expressions of hyaluronidase,hyaluronic acid and CEA which are correlated with invasion and metastasis of cancer. Methods MTT assay was wed to detect the effects of heparin on growth of human breast cancer cell line MDA-MB-231,enzyme linked sorbent assay (ELSA) were employed to detect the expressions of Hyase,HA and CEA of 8 human breast cancer cell lines. Results Heparin had no obvious effect on growth of human breast cancer cell line MDA-MB-231. But it could reduce the expression of hyaluronidasee instead of HA and CEA. Conclusion Heparin is a kind of untoxic medicine and can not directly kill cancer cells, while it can reduce the expressions of some factors correlated with the invasion and metastasis of cancer.
