摘要
目的检测8,24和48 w 龄 SHR 和 WKY 大鼠主动脉平滑肌细胞(ASMC)钙浓度([Ca^(2+)]i)和观察其超微结构。方法采用荧光探针结合计算机图像处理技术和电镜。结果三鼠龄WKY ASMC[Ca^(2+)]i之间没差别(P>0.05)。SHR ASMC[Ca^(2+)]i随鼠龄增加而升高,但8 w 龄 SHR 与同龄 WKY 大鼠ASMC[Ca^(2+)]i 之间没差别(P>0.05)。与 WKY 大鼠相比,24和48 w 龄 SHR ASMC 超微结构异常如异形核,线粒体肿胀和管状结构扩张,但在8 w 龄 SHR 未见这些改变。结论 ASMC[Ca^(2+)]i 升高与成年和老年 SHR 高血压以及 ASMC 超微结构异常有关。
Aim To investigate the ultrastructure of rats vascular muscle cell and its intracellular calci- um concentration at differernt age.Methods In- tracellular calcium([Ca^(2+)]i)and the ultrastructure at aortic smooth muscle cells(ASMC)of 8,24,and 48 w old SHR and WKY rats were examined.Fluores- cent probe combined with digital image processing technique and transmission electron microscope were used.Results There was no significant difference in [Ca^(2+)]i in ASMC of WKY rats at three ages groups(P>0.05).[Ca^(2+)]i in ASMC of SHR was es- calated with the increase of age.[Ca^(2+)]i in ASMC between SHR at 8w old was similar to that of age matched WKY rats(P>0.05).Uhrastructural alter- ations were shown in ASMC of SHR at 24w,and 48 w but not at 8w old.The increase of age were repre- sented as heteromorphous nulei,swelling of mito- chondria,and expansion of tubular systems.Con- clusion Abnormal ultrastructure in ASMC of adult and aged SHR may be ascribed to the increase of [Ca^(2+)]i.
