摘要
利用不同破壁技术对玉米花粉进行破壁处理,采取正交设计对玉米花粉多糖的分离提取条件进行研究。结果表明,花粉破壁处理是影响多糖产量的主要因素,其次为料液比,浸提温度和浸提时间。酶解破壁法和机械破壁法均显著优于温差破壁法,而酶解破壁法与机械破壁法差异不明显。机械破壁法操作方便且成本较低,因此认为适合在多糖提取中使用。玉米花粉多糖分离提取的最优条件为:采取机械破壁法,料液比为1:6,浸提温度为70℃,浸提时间2h。
Corn pollen was processed by different wail-breaking methods before extraction. The orthogonal test design was used in extraction optimization of pollen polysaccharide of corn (PPC). It showed that the pollen wall-breaking process was the main factor influencing PPC extraction, followed by the proportion of pollen to solution, the extraction temperature and extraction time. The enzyme wall-breaking method and machine wall-breaking method were better significantly than temperature-difference wall-breaking method, but there showed no significant differences between the enzyme wall-breaking and machine wall-breaking method. As machine wall-breaking had advantages such as convenient and low cost over enzyme wall-breaking method, it was suitable in PPC extraction. For machine wall-breaking method the optimized conditions for PPC were: 1:6 proportion of pollen to solution and 70℃ of extraction temperature for 2 hours.
