摘要
从抗人脑胶质瘤单克隆抗体杂交瘤细胞中抽提总RNA,并以Oligo(dT)柱纯化mRNA,经逆转录-聚合酶链反应(RT-PCR)直接扩增出318bp的轻链可变区(VL)cDNA片断,经核苷酸序列分析研究,证实已获得了SZ-39单抗轻链可变区基因。这为今后进一步制备基因工程抗体打下基础。
We both have extracted total RNA and purified mRNA by(dT)cellulose in the hybridoma cell of anti-human brain glioma monoclonal antibody and have designed a set of oligonucleotide primers to amplify the cDNA of mouse immunoglobulin light chain variable-region gene by polymerase chain reaction.The primers incorporate restriction sites that allow the cDNA of the variable domain to be force-cloned for sequencing and expression.The result shows that the length of light chain variable-region gene of this antibody is 318 base pairs. This study based for antibody production with genetic engineering.
