摘要
目的:研究了青蒿素与还原型谷胱甘肽之间的相互作用。方法:在含20%乙醇的 B-R 缓冲介质(pH 7.2)中,利用电化学分析方法,对青蒿素与还原型谷胱甘肽的相互作用进行研究。结果:在1.0×10^(-3)mol·L^(-1)青蒿素溶液中,加入谷胱甘肽4.0×10^(-1)mol·L^(-1)时,青蒿素在银电极上-0.64 V 的还原峰消失,于-0.96V 处出现1个新的还原峰,该峰为二者加合物的还原峰,该加合物的形成导致青蒿素的还原峰负移320 mV。通过青蒿素和谷胱甘肽共存体系随 pH 的变化和红外光谱进一步说明了加合物的形成。结论:青蒿素与谷胱甘肽相互作用形成加合物,有可能会影响青蒿素的药理活性。
Objective:To study on the interaction of artemisinin and reductive glutathione. Methods:In Britton - Robinson buffer solution ( pH 7.2) containing: 20% ethanol ( v/v), the interaction of the artemisinin and reductive glutathione was studied by electroanalytical chemistry. Results:The reductive peak of the artemisinin at -0. 64 V disappeared,and a new reductive peak appeared at-0. 96 V in the presence of 4. 0 × 10^-5 mol · L^-1 glutathione. The new reductive peak revealed that adduct of artemisinin and glutathione was formed. The experiments of altering pH of glutathione system and IR spectra analysis were used to confirm the formation of the adduct. Conclusion:The adduct of artemisinin and reductive glutathione was formed. It showed that glutathione can affect the bioactivity of artemisinin.
