摘要
目的建立基因扫描(GeneScan)技术鉴定ABO血型基因型的方法。方法根据O1基因在261位G缺失而非O1基因无缺失,B基因第803位核苷酸与A基因不同的特点,设计4对引物,分别在上游引物5′端标记荧光基团,PCR-SSP扩增后,用基因扫描技术鉴定ABO血型基因型。结果129例样本用GeneScan技术鉴定的ABO血型结果与血清学结果完全符合,A、B、O基因频率分别为0·198、0·159、0·643。其中AA基因型8例(6·2%),AO基因型29例(22·5%),AB基因型6例(4·7%),BB基因型6例(4·7%),BO基因型23例(17·8%),OO基因型57例(44·2%)。结论GeneScan技术鉴定ABO血型基因型具有可行性,提供了一种可选择的方法。
Objective To establish a GeneScan technique to determine ABO blood group genotypes. Methods According to 01 allele sequence having a single G deletion at nucleotide at position 261 comparing to non-O1 allele and B allele differing from A allele by single base substitutions within the coding sequence at nucleotide position 803, four pairs of primers were designed and labeled by fluorescence at 5'terminal of the upper primer. It was used to identify ABO genotypes by use of GeneScan technique after polymerase chain reaction-sequence-specific primer (PCR-SSP). Results The ABO genotypes of 129 samples determined by gene scan technique were accordant to the phenotypes by serological technique. The frequency of A, B, and O were 0. 198, 0. 159 and 0. 643 respectively. AA, AO, AB, BB, BO and OO genotypeswere 8 (6.2%), 29(22.5%), 6(4.7%), 6(4.7%), 23(17.8%), and 57(44.2%) respectively. Conclusion Application of gene scan technique to determine ABO blood group genotypes was feasible and provide another method to identify ABO genotype.
