摘要
目的:观察缺血再灌注大鼠海马神经元超微结构的改变。方法:雄性SD大鼠32只,随机分为缺血再灌注组和假手术组。缺血再灌注组制备缺血再灌注模型,电镜下观察两组大鼠海马神经元超微结构的变化。结果:缺血再灌注可使海马神经元超微结构尤其是线粒体出现明显损伤;缺血再灌注2h出现的神经元水肿、线粒体肿胀等损伤是可逆的;再灌注6h受损神经元增多,细胞器减少,线粒体外膜、线粒体嵴断裂;再灌注12h神经元损伤更为严重,残存的线粒体崩解呈空泡状。结论:缺血再灌注可损伤大鼠海马神经元超微结构,其作用机制可能与线粒体内Ca2+超载、NOS增加等因素有关。
Objective:To observe the ultrastructure changes of hippocampus neurons in ischemia reperfusion rats. Methods: 32 SD rats were divided into ischemia-reperfusion (IR) group and sham-operation group at random. The IR rat model was made in IR group. The uhrastructure changes of hippocampus neurons were observed under electronmicroscope. Results: IR could make obvious damages on hippocampus neurons, especially for mitochondria. The changes of neurons edema and mitochondria swelling in IR 2h group were reversible. In IR 6h group, the damaged neurons increased: the organelle decreased, the adventitia of mitochondria and the crista mitochondriales broke. In IR 12h group, the damages of neurons were more serious: the remaining mitochondria were in vacuole-shape. Conclusions: IR can damage the ultrastructure of hippocampus neurons, the mechanisms may has relationship with overload of Ca^2+ and increasing of NOS in mitochondria.
