摘要
目的探讨肺癌细胞耐药性与凋亡蛋白caspase8、bcl-2、细胞色素C及bcl-2 mRNA异常表达的关系。方法培养肺癌A 549耐药株(A 549R)和敏感株(A 549S),采用逆转录聚合酶链反应技术(RT-PCR)对两株细胞bcl-2 mRNA进行扩增并作琼脂糖电泳,设-βactin为内对照比较其表达水平;W estern B lot法检测caspase8、bcl-2、细胞色素C蛋白表达并比较两株细胞中上述蛋白质表达水平。结果A 549S凋亡启动蛋白caspase8及细胞色素C的表达高于A 549R(P<0.05),而bcl-2蛋白表达低于后者(P<0.05);bcl-2 mRNA表达相对丰度分别为:A 549S 8.74±1.81,A 549R 10.29±2.92,差异无统计学意义(P>0.05)。结论bcl-2的过度表达可能使caspase8和细胞色素C蛋白表达降低从而造成细胞耐药,但bcl-2的这种过度表达可能是在转录或其后的过程而非基因水平发生。
Objective To investigate the relations between the chemotherapy resistance in lung cancer cell (LCC) A549 and the expression of apoptosis protein including caspaseS, bcl-2 and cytochrome C and bcl-2 mRNA. Methods Bcl-2 mRNAs of sensitive A549 cell (A549S) and drug-resistant A549 cell (A549R) cultured were amplified by reverse transcription polymerase chain reaction (RT-PCR), and the expressions in the two strains were compared by AG electrophoresis, with β-actin as control. The caspaseS, bcl-2 and cytochrome C proteins in the two strains were measured and assessed by Western Blot. Results The caspase8 and cytochrome C protein expressions were higher in A459S than in A549R(P〈0. 05). The bcl-2 protein expression was lower in A459S than in A549R (P〈0.05). But there was no significant difference between the level of bcl-2 mRNA expression in strain A549S (8.74±1.81) and that in strainA549R (10. 29±2. 92)(P〉0. 05). Conclusion The upregulationof the bcl-2 protein could depress the caspase8 and cytochrome C protein expression and so might be one of the reasons for drug resistance in lung cancer cell, but the upregulation might happen at the level of translation or thereafter not at the mRNA level.
