摘要
目的探讨糖尿病鼠肾组织损伤的可能机制及格列齐特的保护作用。方法腹腔注射链脲佐菌素建立动物模型,采用RT-PCR法测定各组实验鼠肾组织RAGEmRNA的表达。结果糖尿病模型组实验鼠肾组织RAGEmRNA表达增加,其RAGE/β-actin相对值明显高于对照组(p<0.01)。应用格列齐特治疗后,其肾组织RAGEmRNA表达较模型组显著性降低,其RAGE/β-actin相对值显著低于模型组(p<0.01)。结论RAGEmRNA的高表达可能参与了糖尿病鼠高血糖及糖基化终产物造成的肾组织损伤,而格列齐特可降低RAGEmRNA的表达可能对其介导的糖尿病鼠肾组织损伤有一定的保护作用。
Objective To explore the possible mechanism of renal tissue damage and protective effect of Cliclazide in diabetic rats. Methods Animal model was induced by intra-abdominal injection of streptomtocin. The expression of (Receptor for Advanced Glycation End Products) RAGE mRNA in rat renal tissue was measured by RT-PCR. Results The RAGE mRNA expression was increased in kidney of experimental diabetic rats. RAGE/β- actin ratio was significantly higher than those of the control (P〈0.01). After treatment of Cliclazide, RAGE mRNA expression decreased significantly and RAGE/β-actin ratio was obviously lower than those of the control, Conclusions The high expression of RAGE mRNA may participate in the renal damage of diabetic rats, Cliclazide can protect the renal tissue from the impairment of hyperglycemia and advanced glycation end product,
