从二仙汤拆方对大鼠卵泡颗粒细胞分泌功能的影响探讨方剂组成原理 被引量:16

摘要 目的观察二仙汤及其拆方对大鼠卵泡颗粒细胞分泌雌二醇(estradiol,E_2)的影响,以期阐述二仙汤中各单味药的作用地位和相互间的配伍关系,从细胞水平揭示其配伍组方的原理。方法采用单味药研究法、药物组间关系研究法、撤药研究法等拆方法,用放射免疫法观察二仙汤各拆方直接加药法对颗粒细胞分泌E_2的影响。结果①二仙汤中单味药黄柏、仙茅刺激颗粒细胞分泌E_2的作用大于其他各单味药。②全方去知母组E_2的分泌的作用强于全方组。③淫羊藿与巴戟天的合煎液比其两者分煎液再共同作用于颗粒细胞更能提高E_2水平。结论二仙汤中黄柏和仙茅对颗粒细胞分泌E_2有直接的促进作用。淫羊藿协同作用明显,淫羊藿与巴戟天合煎促进颗粒细胞分泌E_2的作用大于分煎。知母对颗粒细胞分泌E_2有一定的抑制作用。体外实验与体内实验结果的关系有待进一步研究。 OBJECTIVE To investigate and explain the composing mechanism of the formula “Erxian decoction” .METHODS The effects of every herbs in Erxian decoction and its different disassembled prescription(disassembled group removed one drug, disassembled prescription with two kidney wanning herbs or Yin nourishing formula) on the level of estradiol ( E2 ) of granulose cells were measured by RIA. RESULTS ① Xian Mao and Huang Bo in the formula stimulated the secretion of E2 more than other herb in Erxian decoction. ②The disassembled prescription, which removed Zhi Mu, had a better effect than the whole formula on the secretion of E2. ③The stimulating effect on the E2 secretion of mixed decoction of Ying Yang Huo and Ba Ji Tian was better than that of their separated decoction. CONCLUSION Huang Bo and Yin Yang Huo were the most important parts in Erxian decoction for stimulating E2 secretion. Zhi Mu inhibited the secretion of E2. The separated decoction of Ying Yang Huo and Ba Ji Tian has slight stimulating effect on the E2 secretion, compared with their mixed decoction.
