籽粒硬度Pinb基因反义表达载体构建及遗传转化

Construction and Wheat Genetic Transformation of Antisense Expression Vector Harboring Pinb Genes

摘要 籽粒硬度是小麦品质改良的重要目标性状。利用含有Ubi启动子、Bar表达盒的基础质粒pAHC25,构建了含有Pinb基因的植物反义表达载体pAHantipB,其中pAHantipB载有小麦籽粒硬度Pinb基因的反向序列,可用于小麦遗传转化,利用花粉管通道法将其转入软质小麦品种京411,获得1株转基因植株,T1代转基因植株抗性筛选及PCR检测结果表明,转化率为0.17%。基因组DNA斑点杂交证实了外源基因在受体小麦基因组中的整合。 Grain hardness is one of the most important characters in wheat quality improvement. Here, Antisense expression vector pAHantipA, harboring the reverse sequence of Pinb , respectively, were constructed using pAHC25, as basic plasmids, which harbors the ubiquitin promoter and selection gene Bar , which can be used in wheat genetic transformation. A transgenic plant with antisense Pinb was obtained by pollen-tube pathway with Jing 411 as receiptor. Basta resistance test and PCR analysis indicated that the transgenic plants with foreign Pinb gene were obtained with the efficiency of 0. 17%, respectively. Southern dot blot analysis further confirmed the result.
